When I began working on this, it rapidly became clear that Evelyn Duke Brandenberger had made wonderful contributions to Duke research, but had allowed her fondness for the Duke-Byrd connection to push her out considerably in front of her data. For example, it is often repeated that Wm. Byrd took William Duke on his survey of the VA/NC line, when in fact Byrd's diary (both the public and private versions) has been in print for decades and clearly shows that it not true. Basically the documentary evidence for the connection did not add up, but it was the yDNA evidence that put the final nails in the coffin of this hypothesis. The yDNA study is reported at:

If you go to the "Earliest Documented Ancestors" link at the bottom of the homepage, you'll see that in Group 2 that William Duke of Brunswick Co VA and Warren Co NC is an excellent yDNA match for individuals whom we know to be descended from John Duke of Isle of Wight. The choice of William Duke and the younger John Duke of Isle of Wight to live next door to one another in both VA and NC, and not next door to any of the other Duke families, had always seemed to me inadequately explained by Mrs. Brandenberger, who placed William in the Duke-Byrd family instead of the family of John Duke of Isle of Wight. To be brutally frank, Mrs. Brandenberger did not want to be descended from a Duke family known to have had a member (Bridget Duke, mother of the younger John) on the parish charity rolls, but instead preferred the socially prominent Duke-Byrd connection. Many have been tempted to see their data through lenses like this, her mistakes are certainly not unique to her. In any case, Group 4 in the DNA study is actually the group descended from the Col. Duke/Capt. Henry Duke line, and is also the line associated with the famous Duke family of NC that founded American Tobacco Company. A few members of Group 4 may be "accidental" matches within a very common yDNA haplogroup, but the core of the family is pretty clearly the Col. Duke line. If William Duke of VA and NC had been a son of James Duke and Mary Byrd, his descendants should fall in this Group 4, rather than Group 2.
